New Theophylline Derivatives with Potential Pharmacological Activity

Theophylline, a methylxanthine compound, is known as an efficient bronchodilatator drug, having also anti-inflammatory and immunomodulatory effects. In order to improve its pharmacological profile and to reduce also the serious side effects that appear at high concentrations, new theophylline derivatives have been synthesized. The new compounds are obtained in two steps by the reaction of 8-substituted-theophyllines with epoxy-propyl-acetaminophen. The chemical structure of the synthesized compounds has been elucidated by their H-NMR spectra. The potential bronchodilatator effects of the synthesized compounds have been also established. Rezumat Teofilina, un compus metilxantinic, este cunoscută ca un eficient medicament bronhodilatator, având totodată şi efecte antiinflamatoare şi imunomodulatoare. În scopul îmbunătăţirii profilului său farmacologic şi pentru a reduce serioasele efecte secundare ce apar la doze mari, au fost sintetizaţi noi derivaţi de teofilină. Noii compuşi, nemenţionaţi în literatura de specialitate, au fost obţinuţi în două etape, prin reacţia derivaţilor de teofilină substituită în poziţia 8 cu diferiţi radicali cu epoxi-propil-paracetamol. Structura chimică a compuşilor sintetizaţi a fost confirmată prin spectroscopie H-RMN. Compuşii au fost testaţi şi pentru evidenţierea potenţialelor efecte bronhodilatatore.
